Brain vs axis
PT-141 is a melanocortin (MC4) receptor agonist — a cousin of Melanotan 2 — acting on desire circuits in the hypothalamus regardless of hormone levels. Kisspeptin is the master upstream signal for GnRH release; its effect on libido in human studies appears to run through both hormone elevation and limbic-brain activation seen on fMRI.
Dosing and timing
PT-141: 1–2 mg subcutaneous about 45 minutes to 4 hours before, no more than once in 24 hours. Kisspeptin-10: 50–100 mcg subcutaneous, often 2–3× weekly, with effects assessed over weeks via hormone labs rather than the same night.
Side effects
PT-141 causes nausea in roughly 40% of users at 1.75 mg, plus flushing and transient blood-pressure rises — it is contraindicated with uncontrolled hypertension. Kisspeptin-10 is well tolerated in trials, with the caveat that continuous exposure can desensitize the GnRH system, so pulsatile dosing matters.
Can you stack them?
Rarely stacked with each other. Kisspeptin-10 appears alongside gonadorelin or in TRT-adjunct discussions; PT-141 is sometimes combined at low dose with Melanotan 2, though both hit melanocortin receptors and nausea compounds.
